Development and Validation of a Procedure for Quantitative Determination of Flavonoids in Sea Buckthorn Leaves by Spectrophotometry

Sea buckthorn fruits are a pharmacopoeial raw material used to produce sea oil. Other parts of the plant (bark, leaves, and shoots) objects research aiming expand base use limited natural resources rationally. leaves rich in biologically active compounds, including flavonoids. The aim study was develop validate an analytical procedure for quantification flavonoids using spectrophotometry visible spectral region. Materials methods. focused on dried harvested Voronezh region 2022. These were collected from wild plants at three phenological stages: fruit setting (I), single ripening (II), mass (III). To quantify authors based measuring absorbance solution containing products reaction between aluminium chloride. Results. absorption maximum complex alcohol extract chloride observed 402±2 nm; wave-length corresponds luteolin. optimal extraction conditions included 70% ethanol as solvent, solvent ratio 1:150, particle size 0.5 mm, cycle 45 min. Conclusions. developed validated spectrophotometric total flavonoids, expressed luteolin, leaves. can be standardise this herbal drug. As study, flavonoid content highest (up 1.5%) stage I (collected mid-June) gradually decreased through mid-July end August. Since is source oil early collection may decrease its value or lead non-compliance with requirements, recommended time harvesting should coincide that fruits.
